Fumonisins are mycotoxins produced by the fungus Fusarium moniliforme. Frequent, almost universal inhabitants of corn, fumonisin B1 and fumonisin B2 are present in most corn samples tested, often at greater than 1 ppm. Fumonisin is thought to cause equine leukoencephalomalacia in horses, swine pulmonary edema, and human esophageal cancer. T he FDA/USDA Working Group on Fumonisin advises levels of less than 4 ppm for products for human consumption, less than 5 ppm in horse feed, less than 20 ppm for swine feed and less than 30 ppm for cattle feed. T he European Union has provided the following recommendations:
  • Maize-based breakfast cereal and snacks 0.8 ppm
  • Maize grits/flour/meal and refined maize semolina 1 ppm
  • Other maize-based foods for direct human consumption 1 ppm
  • Processed maize-based foods for infants and young children and baby food 0.2 ppm
  • Unprocessed maize not intended for wet milling 4 ppm

  • Grind and weigh sample
  • Add methanol/water mixture

Dilute and Filter

  • Dilute extract

  • Pass filtrate over OchraTest affinity column
  • Wash column with buffer and water
  • Elute ochratoxin and collect in cuvette

Measure

  • Add developers and place cuvette into a calibrated fluorometer, or
  • Inject eluate into HPLC
  • Determine fumonisin concentration
